Uptake and esterification of palmitrate by rat diaphragm in vitro.

چکیده
The contribution of exogenously supplied palmitate to the intracellular palmitate pool and its role in esterification were studied for the intact rat diaphragm in vitro. Palmitate-1-(14)C attached to albumin in various molar ratios (nu) was taken up by the tissue in an initial rapid phase which led, after 4 min, to a steady-state level of tissue free fatty acid. The level was determined by nu but also by the albumin concentration below 2 g/100 ml. The exogenous palmitate taken up constitutes one-quarter to three-quarters of the intracellular palmitate pool as nu is increased from 0.5 to 5.5 (albumin concentration 2 g/100 ml). Turnover rates of intracellular palmitate at a given nu are stable over 35 min of incubation, as indicated by constant rates of esterification (largely to triglyceride, but also to phospholipids) and by the unchanging specific activity ratios between intracellular and medium palmitate-1-(14)C. These ratios also indicate that only a portion of the palmitate taken up is available for exchange with the albumin complex in the medium. Calculation of esterification rates at various nu values suggests that not all of the palmitate is in a pool that is available for esterification.

Effect of pituitary extracts upon glucose uptake by diaphragms from normal, hypophysectomized, and hypophysectomized-adrenalectomized rats.
The measurement of the glucose uptake of rat diaphragm muscle in vitro is of interest because striated muscle is quantitatively the most important tissue for glucose disposal in the animal body and because glucose uptake, for which the hexokinase reaction is presumably the rate-determining enzymatic step, has been shown to be under hormonal control. The evidence for hormonal control obtained so...

The effect of increasing the intracellular pool of monoolein upon the subsequent uptake and esterification of oleic acid was investigated using in vitro rat jejunal slice techniques. The mucosal pool of monoglyceride was expanded by preincubation of jejunal slices in a monoglyceride-containing bile salt medium at a temperature close to 1 degrees C, which inhibited esterification. Subsequent inc...
متن کاملEffect of Insulin on Carbohydrate Metabolism of Skeletal Muscle Fibers and Diaphragm from Control and Pancreatectomized Rats*Jf
Rat diaphragm muscle has been used almost exclusively for the investigation in vitro of skeletal muscle metabolism. However, the diaphragm is a specialized muscle adapted to constant activity and, with respect to oxygen consumption (1) and oxidative metabolism, seems to occupy an intermediate position between constantly active heart muscle and intermittently active skeletal muscle (2, 3). Histo...

There is abundant evidence that pituitary and adrenal cortical hormones influence insulin so as to produce marked alterations in its customary action on tissue metabolic patterns. There is little doubt that this reciprocal influence is manifest in many types of tissue and is particularly well established in the cases of liver and muscle. The injection of pituitary or adrenal hormones into norma...

The interaction between long-chain and medium-chain lipids during intestinal absorption was examined using several model systems. A decrease in steady-state triolein (LCT) output in thoracic duct lymph after addition of trioctanoin (MCT) to the duodenal infusion confirmed previous studies in unanesthetized rats which demonstrated inhibition of steady-state LCT uptake from the small intestinal l...
